NMN is an oral NAD+ precursor taken to counter the age-related decline in cellular NAD+. It is a vitamin-B3-adjacent metabolite, not a hormone or anabolic. It does not raise testosterone or IGF-1 or build muscle. Human trials show it reliably raises blood NAD+ but the downstream healthspan/performance benefits are modest and still being established. Rated 1 as a cellular-health/longevity supplement with no meaningful performance-enhancing effect.
Overview
One of the two most popular oral NAD+ precursors (alongside NR). NMN sits one step closer to NAD+ than nicotinamide riboside in the salvage pathway and is taken to restore declining age-related NAD+ levels for longevity and cellular-energy goals. Purpose & Use Cases
NAD+ Restoration
Raises circulating NAD+, roughly a two- to two-and-a-half-fold increase in whole blood within the first few weeks in human trials, tapering somewhat with continued dosing, to offset the age-related decline.
Longevity / Sirtuin Support
Provides NAD+ substrate for sirtuins, the deacetylases central to many longevity hypotheses. Frequently stacked with resveratrol (a sirtuin activator) on a "fuel + activator" rationale.
Metabolic / Physical Function
Small RCTs report modest improvements in aerobic capacity in amateur runners and some muscle-function measures in older adults, encouraging but early, and not consistently muscle-building.

NMN is one step from NAD+ in the salvage pathway and raises NAD without the flushing niacin causes. It is used for the same age-related NAD-decline rationale as the NAD+ page.
TMG is optional here, not mandatory
The idea that you must take TMG with NMN to protect methylation is largely theoretical at supplement doses, the one NR trial that measured it (3,000mg/day) found only a mild homocysteine rise with methylation markers otherwise preserved. TMG is cheap, low-risk insurance, but the "mandatory" framing is overstated for NMN/NR.
US legal status: mostly resolved, but not bulletproof
The FDA excluded NMN from the dietary-supplement definition in 2022, then reversed that position in September 2025 after an industry lawsuit, confirming beta-NMN is not excluded. It still requires NDI notification and a future administration could revisit the ruling, not a doping issue, a supplement-regulation one.

Human trials have tested doses from ~100mg up to 2,000mg/day (e.g. 1,000mg twice daily for 14 days in one trial); the 300-1,200mg/day range has the most repeated safety data (e.g. 300/600/1,200mg/day for 6 weeks in amateur runners with no reported adverse events). NAD+ rises within 2-4 weeks. There is no strong evidence that megadosing produces proportionally more benefit. Unlike nicotinic acid (niacin), NMN does not cause flushing. Some prefer sublingual/liposomal forms for absorption, though oral NMN already raises blood NAD+ effectively.
NMN is rapidly metabolised, so its effect is measured by the sustained rise in blood NAD+ rather than a parent-compound half-life.

General Mitigation Strategies
Very well tolerated in human trials up to ~1,200mg/day with no significant adverse effects. Any minor GI effects are usually resolved by taking it with food or lowering the dose. Note the main open question with NMN is efficacy/regulatory status, not safety.

TMG (Trimethylglycine / Betaine)
Methyl-donor "insurance." NAD turnover produces nicotinamide, which the enzyme NNMT clears by methylation using SAM, spending methyl groups and generating homocysteine. TMG helps remethylate homocysteine back to methionine, buffering that drain.

- Honest evidence: for NMN specifically the methyl-depletion concern is largely THEORETICAL at normal supplement doses. The mechanism is textbook, but it is best-supported for high-dose nicotinic acid (niacin), not NMN/NR, the one NR RCT that actually measured this (NR-SAFE, 3,000mg/day) found only a mild homocysteine rise with the SAM/SAH methylation ratio otherwise preserved, not the meaningful depletion the theory predicts. TMG is cheap, low-risk, and independently lowers homocysteine, so it is reasonable insurance rather than a biochemical necessity for NMN.

How It Works
NMN is converted to NAD+ via the salvage pathway: NMN adenylyltransferases (NMNATs) add an adenylyl group to form NAD+ directly. NAD+ is an essential coenzyme for energy metabolism (glycolysis, TCA cycle, oxidative phosphorylation) and a substrate for sirtuins (SIRT1-7) and PARPs involved in DNA repair and stress response. There has been debate about how NMN enters cells (the transporter Slc12a8 was proposed); in practice, oral NMN reliably raises circulating NAD+ regardless of the exact uptake route.
